چکیده

Monoamine transporters are the main targets of methamphetamine (METH). Recently, we showed that fluoxetine, a selective serotonin reuptake inhibitor (SSRI), decreased METH conditioned place preference (CPP), suggesting that serotonin transporter (SERT) inhibition reduces the rewarding effects of METH. To further test this hypothesis, in the present study we investigated the effects of additional SSRIs, paroxetine and fluvoxamine, on METH CPP in C57BL/6J mice. In the CPP test, pretreatment with 20 mg/kg paroxetine abolished the CPP for METH, whereas pretreatment with 100 mg/kg fluvoxamine prior to administration of METH failed to inhibit METH CPP. These results suggest that paroxetine, a medication widely used to treat depression, may be a useful tool for treating METH dependence. Further, these data suggest that molecules other than the SERT [such as G protein-activated inwardly rectifying K+ (GIRK) channels] whose activities are modulated by paroxetine and fluoxetine, but not by fluvoxamine, are involved in reducing METH CPP by paroxetine and fluoxetine.

INVOLVEMENT OF THE SEROTONIN SYSTEM IN SSRI-INDUCED ANTINOCICEPTION

Serotonin specific reuptake inhibitors (SSRI) may induce antinociception however, the mechanism of this effect is not clear. SSRls increase 5-HT levels in neuronal synapses and facilitate serotonergic activity. In this study, therefore, the activity of para-chlorophenylalanine (pCPA), which reduces 5-HT release, and 5- hydroxy tryptophan (5-HTP), a precursor of 5-HT, were examined on the a...

Fluoxetine as a potential pharmacotherapy for methamphetamine dependence: studies in mice.

The monoamine transporters are the main targets of psychostimulant drugs, including methamphetamine (METH) and cocaine. Interestingly, the rewarding effects of cocaine are retained in dopamine transporter (DAT) knockout (KO) mice, while serotonin transporter (SERT) and DAT double KO mice do not exhibit conditioned place preference (CPP) to cocaine. These data suggest that SERT inhibition decrea...

Guidelines for choice of selective serotonin reuptake inhibitor in depressive illness

The selective serotonin reuptake inhibitors (SSRIs) are the most extensively studied of the newer antidepressants and are increasingly being used as firstline treatment for depression (Anderson et al, 2000). In this article we concentrate on issues that need to be taken into account when selecting one of the five SSRIs marketed in the UK (citalopram, fluoxetine, fluvoxamine, paroxetine and sert...

Background and Objective: Increased level of dopamine in accumbens nucleus has a key role in the rewarding effects or positive reinforcement of abused drugs, whereas serotonin facilitates dopamine release in brain .The aim of this study was to investigate the effect of concurrent use of amantadine and paroxetine on reinforcing effect of morphine in conditioned place preference model in mice. Effects of chronic antidepressant treatments on serotonin transporter function, density, and mRNA level.

To investigate functional changes in the brain serotonin transporter (SERT) after chronic antidepressant treatment, several techniques were used to assess SERT activity, density, or its mRNA content. Rats were treated by osmotic minipump for 21 d with the selective serotonin reuptake inhibitors (SSRIs) paroxetine or sertraline, the selective norepinephrine reuptake inhibitor desipramine (DMI), ...
